In recent five years, in order to solve the key scientific issues of early warning and early diagnosis of gastric cancer, he carried out the screening of candidates for gastric cancer related markers, the principle of new technologies of nanometer detection, the development of multifunctional gastric carcinoma-targeted Nano-Probes, controllable preparation of nano-particles and study on application of nanometer detection devices. His main scholarly contributions are as follows:
1. screening and identification of markers for gastric cancer
He demonstrated that the Phospholipase c, Epsilon 1 (PLC E1) is low or no expression in lesions of atrophic gastritis, and high-level expression in gastric carcinoma. It’s an important molecular marker and can be used to distinguish early gastric cancer and atrophic gastritis. The papers published in PLos One 2012;e47563. He filterd out some respiratory gas markers that exist only in patients with gastric cancer though collecting more than 200 cases of respiratory gas of patients with gastric carcinoma and precancerous lesions. He conducted the hyper-sensitive method of breathing gases electrochemical sensor detection and applied for an international patent (PCT/CN2012/000083). The paper published in Theranostics 2013, Nanoscale Research Letters 2012;7:475.
2. Establishment of 4 principles of nano-probing technology
In order to improve the sensitivity and specificity blood markers of gastric cancer, he firstly reported the principle of hyper-sensitive technology of detecting nucleic acid and Antigen based on self-assembled quantum dots and carbon nanotubes. The oligonucleotide concentration that can be detected is 0.2pM, and Antigen concentration is 0.01nM, and the sensitivity and specificity is over ELISA methods. The multicomponent simultaneous detection can be realized because of different colors depended on the quantum dot size. The paper published in Analytical Chemistry (2008;80:7996-8001). Nature Nanotechnology listed it as Research Highlight and commented it on column of Top Down and Bottom Up in the 2008 period. It says “carbon nanotubes combined quantum dots can be highly sensitive to detect DNA and Antigen” “this system can be extended to biosensor applications”. He firstly reported the phenomena that quantum dots modified by mercaptoacetic acid can enhance the specificity of PCR and the principles of quantitative PCR method based on quantum dot fluorescence quenching, and applied the patents(ZL200610026091.1), which is more significant for testing of micro nucleic acid. He firstly reported the pollution-free and controlled synthesis of nano silver based bacteria as the template which is linked with Raman molecular whose detection signals can be increase 1011 times. The paper is published in Green Chemistry (2010;12:2038-2042). In order to detect out minor amount of maker in saliva, he conduct a method to detect out the microscale antibody in saliva by magnetic Nano particle mission clusters which combines fluorescent quenching fight gene probe and double features protein that can combine Fc fragments of antibody and has the features of MboI enzyme cutting. The method’s sensitivity can reached 0.1ng/mL which is significant to the detection of early and few maker of AIDS HIV, hepatitis virus, and tumor. The paper is published in Analyst (2011; 136:679-683). He established the principles of quantitative PCR technology based on the fluorescence quenching of quantum dots and achieve the invention patents (ZL200610026091.1) which transferred to the Shanghai bio-pharmaceutical company and developed a series of kits.
3. the design and preparation of 3 types of Microfluidic magnetic sensors
In order to capture of gastric cancer cells in the circulating blood, he designed magnetic nanoparticle probes coated by Chitosan and modified by RGD , which is fixed on the surface of Microfluidic channels. The probes achieve the rapid acquisition and quantitative detection of gastric cancer cells in the blood using giant magnetoimpedance (GMI) effect. For cancer-related viruses such as HPV viruses, he designed the GMI sensor micro-fluidic chip system which achieved rapid genotyping of 16/18 HPV virus. For HBV virus, he designed a giant magneto-resistive (GMR) chip sensor which can detect 8 indicators at the same time by using magnetic nanoparticles clusters and combined with linear probes and PCR techniques. The probes achieved rapid genotyping of HBV. The papers are published in Lab-on, respectively a Chip (2012:12 (4): 741-745), Biosensors&Bioelectronics (2011;26:3246-3253), Applied Physics Letters (2010;97 (043702)).
 4. Develop 4 gastric carcinoma-targeted image multifunctional Nano-Probes
In order to achieve the positioning quantitative Visualization of early gastric cancer, he prepared fluorescent magnetic Nano particle probe combined with BRCAA1 (AF208045) antibody which has autonomous intellectual property. The probe can actively target to 5mm diameter gastric cancer of nude rat model, and achieved fluorescent imaging and MRI of gastric cancer. The paper was published in Journal of Nanobiotechnology (2011;9:93), and he was invented to reported in fifth session European clinical Nano medical Conference. He firstly reported the new method of controlled synthesis of up-conversion fluorescent magnetic Nano particle by using double phase system including oil acid and ion liquid. The probes can achieve CT ,MRI and near infrared fluorescent imaging to the gastric cancer model. The up-conversion Nano particle(NaYF4:Yb,Er) modified by cribellate Silicon can achieve targeted therapy and near infrared fluorescent imaging of cancer, carrying tumor chemotherapy drug, under outside magnetic field.
